Aplikasi Elektroforasi Sperma untuk Produksi Ikan Lele Mutiara (Clarias sp.) Transgenik

Abstrak


Penelitian transfer gen hormon pertumbuhan pada sperma ikan menggunakan teknik elektroforasi untuk perbaikan pertumbuhan telah berhasil dilakukan pada spesies salmon, catfish, common carp maupun tilapia. Pemberian kejutan elektrik (125 V/cm dan 50 V/cm dengan jumlah pulsa 3 dan 5) pada sperma ikan lele mutiara memberikan peluang besar gen GH lele dumbo masuk dan bergabung dengan DNA genom ikan inang saat proses elektroforasi. Rekombinasi GH eksogen (GH lele dumbo) dan GH endogen menyebabkan over ekspresi pertumbuhan ikan lele mutiara transgenik. Hasil penelitian menunjukkan rata-rata kelipatan tumbuh ikan lele  mutiara transgenik hasil elektroforasi 125 V/cm-pulsa 3, 125 V/cm-pulsa 5, 50 V/cm-pulsa 3 dan 50 V/cmpulsa 5 berturut- turut 3,53 kali; 3,05 kali; 2,75 kali dan 2,76 kali dari ukuran ikan non transgenik.
