Efek α-Mangostin terhadap Aktivitas NF-kB, Dan Kaspase-3 Enterosit pada Bayi Tikus Model Enterokolitis Nekrotikans

Abstract

Necrotizing enterocolitis (NEC) is a devastating gastrointestinal emergency affecting premature infants  with high  morbidities and  mortalities. Multifactorial combination produced intestinal mucosal injury culminating inflammation, oxidative stress and apoptosis through tumor necrosis factor (TNF)-α, nuclear factor-kB (NF-kB) and caspase-3  activities. Garcinia mangostana called mangosteen  exhibit  antioxidant and anti-inflammatory activities. This study was done to determine mangosteen  effects  in reducing, nuclear factor-kB (NF-kB), and caspase-3 activities  in neonatal rat model of  NEC. Experimental study with complete randomized design was conducted from July to September 2013 in Faculty of Veterinary Medicine, Bogor Agricultural University in Bogor, done in rat model induced with ischemic-reperfusion, enteral  feeding and cold stress. The neonatal Wistar rat were gavaged with α-mangosteen 25 mg/kg bw and 50 mg/kg bw one hour before NEC induction. Intestinal tissue was assessed within 6, 24, 48 and 72 hours to determine NF-kB and caspase-3 reducing activities by immunohistochemical staining. Statistical analysis was done by using Kruskal-Walis,and  pos hoc non parametric Wilcoxon  Mann-Whitney. Comparison  of  median  NF-kB  histoscore showed significant differences between treated groups at hour72 observation. There were also significant differences in median histocore of NF-kB between control positive group (P1) and α-mangosteen 25 mg/kg bw group (P2) and  between P1 group and α- mangosteen 50 mg/kg bw (P3) group but no significant difference between P2 and P3 group at hour 72 observation. Median histoscore of caspase-3 between treated group  showed  significant difference at 72 hour observation. The median oomparison of caspase-3 showed significant difference at 72 hour observation  between control  positive (P1) group and   α-mangosteen 25 mg/kg bw group (P2) and between P1 and  α-mangosteen 50 mg/kg bw (P3) group but not between P2 and P3 group activities. α-mangosteen per oral showed decreased NF-kB, caspase-3 activities in enterocytes in72 hours observation which means  α-mangosteen per oral confirmed tke antiapoptotic response in time dependent manner.
