PENGARUH LIKOPEN TERHADAP KUALITAS DAN KADAR MALONDIALDEHID SPERMATOZOA YANG DIPAJANKAN PADA ZALIR PERITONEUM WANITA DENGAN ENDOMETRIOSIS

Abstract

The quality of spermatozoa should be optimized in order to survive in the peritoneal fluid environment, tubes and uterus, especially in women with endometriosis in which its peritoneal fluid containing oxidant and pro - inflammatory substances that can damage the sperm. Lycopene is a compound that has a high antioxidant activity. Method of lycopene in a series of incubation of sperm preparation ensure an application of new methods that can produce spermatozoa that are more resistant to oxidative stress and inflammatory factors that exist in the peritoneal fluid, tubal and uterine cavity, especially in women with endometriosis . This study aimed to examine the benefits of the addition of lycopene in preparation in protecting sperm's morphology, motility, viability, DNA fragmentation and level of MDA as the result of oxidative and inflammatory peritoneal fluid of women with endometriosis so that sperm quality is maintained properly in intrauterine insemination program . The purpose of this study was to test the effect of incubation of lycopene on the motility of spermatozoa preparation, viability, DNA fragmentation and level of MDA of spermatozoa exposed peritoneal fluid of women with endometriosis. This study was interventional experimental design conducted in the laboratory (in vitro) with spermatozoa as a research subject. The study was conducted at Aster Clinic, Dr. Hasan Sadikin Hospital / Faculty of Medicine, Padjadjaran University-Bandung from March to November 2013. Fifteen sperm samples were prepared and treated with three different methods. The first treatment group was incubated with 5μM lycopene for 30 minutes and then exposed to peritoneal fluid of endometriosis. The second treatment group was given the peritoneal fluid of endometriosis. While the third group was given the sperm wash medium. Examination of viability, motility, morphology, DNA fragmentation using the SCA method, whereas MDA levels using colorimetric method. Analysis was performed on four times, 0, 3, 6, and 24 hours. As the results, the viability of spermatozoa in group one which was treated by lycopene incubated in the peritoneal fluid of women with endometriosis was higher than without lycopene. There was no different in motility between group one and two. The morphology of spermatozoa in group one was maintained significantly better than group two and three.  There was no statistical different in DNA fragmentation between all groups. The MDA levels in group one was significantly lower than other groups. In conclusion, lycopene use in the sperm preparation is proven to be useful in maintaining quality of spermatozoa, especially its viability and morphology, as well as lowering MDA levels.
