PEMBENTUKAN TANAMAN PADI HAPLOID GANDA TAHAN PENYAKIT HAWAR DAUN BAKTERI (HDB) dan TOLERAN KERACUNAN Fe MENGGUNAKAN PADI LOKAL INDONESIA

The accessions of Indonesia local rice germplasm which has long been cultivated by farmers has the potential alleles that can be utilized. Some local rice germplasm accessions was selected based on allele mining research is Parekaligolara (Indica, IRN 1541) with known resistance genes to pathogenic allele Bacterial Leaf Blight (BLB), Xa7 and Markuti (Indica, IRN: 5754) which has poisoned tolerant gene alleles Fe, OsIRT. Germplasm has been used as genetic material in a double cross IR54/Parekaligolara//Bio110/Markuti. The formation strains the hope new rice tolerant to biotic (BLB disease resistance) and abiotic (Fe toxicity tolerance). The purpose of this study was to obtain dauble haploid population of rice that has resistance gene of bacterial leaf blight and Fe toxicity tolerance throught anthera culture method and SSR molecular marker selection test. Formation of double haploid population for BLB resistance throught anther culture technique with SSR molecular marker selection. Marker are used for BLB resistance is : RM20589, RM20590, RM20591, Xa7-LD34 dan Xa7-LD40, and tolerant to Fe toxicity is OsIRT1 and OsIRT2. BLB disease resistant trait is controlled by more than one dominant gene. Where as the genotype performance showed that the marker Xa7-LD40 as co-segregation marker with resistance trait. In the formation of double haploid plants BLB resistance of 50 plants tested showed 10 resistance plants, 25 medium resistance plants, 15 susceptible plant. Double haploid plants are tolerant to Fe toxicity there are 25 plants resistant, 12 plants medium tolerant and 13 plants sensitive.
