Combination of EH-MSCs, Glutathione, and Vitamin C: Its Effect on IL 10 and CD-163 Gene Expression in Hyperpigmentation Model Mice

Abstrak


Hyperpigmentation of the skin is the result of UVB exposure which causes oxidative stress due to increased ROS, causing a number of skin problems including melanin accumulation. Exosomes can affect melanocyte activity, Exosomes as small vesicles released by cells, can affect melanocyte activity and play an important role in various hyperpigmentation processes. The study aims to determine the effect of exosome mesenchymal stem cell hypoxia (EH-MSC) and glutathione with vitamin C on IL-10 and CD163 gene expression. Thirty (30) mice were used, randomly divided into 5 treatment groups, namely K1 (group 1) healthy mice, K2 (group 2) control without intervention, K3 (group 3) glutathione injection and vitamin C treatment, K4 (group 4) exosome treatment, and K5 (group 5) exosome injection and glutathione and vitamin C treatment. The result revealed that the highest IL-10 gene expression was observed in the group treated with treatment of EH-MSCs alone (K4) compared to the control group. Meanwhile, the highest CD-163 gene expression results were found in the group with combination of EH-MSCs and glutathione with vitamin C treatment (K5). These findings demonstrate that the administration of EH-MSC and glutathione with vitamin C increasing the expression of IL-10 and CD163 in C57BL / 6 mice with UVB-induced hyperpigmentation model.
