EFEK ESTROGENIK HERBISIDA PARAKUAT TERHADAP DIFERENSIASI KELAMIN EMBRIO PENYU HIJAU
Abstrak
To detect the effect of paraquat dichloride to disrupt sex differentiation of green turtle (Chelonia mydas) embryos were examined. Eggs from the green turtle were incubated under temperature of 26 ± 0,5oC and sprayed with paraquat concentration of 58, 125, 270, and 580 mg/kg from day 24 (Stage 22) until day 60 (Stage 29) every two days. Aquades were sprayed in control groups. The gonad was isolated then fixed in Bouin's solution to make histological preparation by paraffin method and haematoxilin-eosin staining. The slice were observed under 400x and 1000x magnification of compound microscope. The result indicated that paraquat dichlorides were highly significant to viability (P £ 0.01) decreasing and significantly influent to sex differentiation (P £ 0.05) which signated by increasing percentage of gonadal intersex of embryos. Hence, paraquat dichloride is capable to increase 71,43% number intersex embryos.
