PENGARUH BENZIL ADENIN DAN TIDIAZURON TERHADAP MULTIPLIKASI TUNAS PIRETRUM (Crysanthemum cinerariifolium (Trevir.) Vis.) KLON PRAU 6 SECARA IN VITRO
Abstrak
Phyretrum is one of the botanical pesticides to be further developed for subtitutions of synthetic pesticides phyretriod which is found to be harmful both to the environment and human being. An experiment on the effect of cytokinin BA and TDZ on in vitro shoot multiplication of piretrum clone Prau 6 was performed and arranged in Split-Plot Design with 10 replication. The main plot was explant source i.e. MS + BA 3,0 mg l-1- derived shoot or MS + TDZ 3,0 mg l-1 ones. The sub plot consist of 11 combinations of cytokinins BA and TDZ, as a single or combination of both BA and TDZ : 3,0 mg l-1 BA; 3,0 mg l-1 TDZ; 0,1 mg l-1 BA + 0,1 mg l-1 TDZ; 0,2 mg l-1 BA + 0,2 mg l-1 TDZ; 0,3 mg l-1 BA + 0,3 mg l-1 TDZ; 1,0 mg l-1 BA + 0,1 mg l-1 TDZ; 2,0 mg l-1 BA + 0,2 mg l-1 TDZ; 3,0 mg l-1 BA + 0,3 mg l-1 TDZ; 0,1 mg l-1 BA + 1,0 mg l-1 TDZ; 0,2 mg l-1 BA + 2,0 mg l-1 TDZ; and 0,3 mg l-1 BA + 3,0 mg l-1 TDZ. Parameters, observed were the period of shoots initiation, number of shoots, shoots height, percentage of the grown explant and the number of proliferated shoots, at 4 week after culture. The resulst showed that explant sources and cytokinin applied significantly affected to the number of shoots and height. The shoots derived from MS + BA 3 mg l-1 media resulted in the highest number of shoots (7,6) after subcultured into MS + BA 0,3 mg l-1 + TDZ 3 mg l-1 media, with 1,6 cm of plant height.
