ABSTRAK

Pendahuluan: Banyak bahan kedokteran gigi yang beredar diimpor dari luar negeri dan dijual bebas di pasaran. Hal ini sempat menimbulkan kegelisahan bagi para muslim yang mencari perawatan gigi yang halal. Status halal bahan yang digunakan untuk perawatan gigi masih belum jelas. Penelitian ini bertujuan untuk mengidentifikasi DNA porcine pada bahan mahkota sementara, bahan basis gigi tiruan, dan bahan cetak hydrocolloid irreversible menggunakan Real-Time PCR. Metode: Jenis penelitian menggunakan penelitian deskriptif,  metode quota sampling digunakan untuk pengambilan sampel. Tiga bahan mahkota sementara, tiga bahan basis gigi tiruan dan tiga bahan cetak hydrocolloid irreversible dengan merk yang berbeda digunakan sebagai sampel. Penelitian ini dilakukan dalam dua tahap. Pertama, ekstraksi DNA pada sampel dan kedua dan dilakukan analisis Real-Time PCR berdasarkan kurva amplifikasi dan skor Ct pada channel yellow dan channel green. Hasil: Analisis sampel berdasarkan channel green menunjukkan bahwa semua bahan tidak mengandung DNA porcine, tetapi empat dari sembilan sampel mengandung DNA vertebrata. Simpulan: Semua jenis bahan yang diuji tidak satupun yang mengandung DNA porcine.

ABSTRACT

Introduction: Most of dental materials circulates are imported from abroad and sold freely on the market. It cause an anxiety for Muslim community who looking for halal dental treatment. Halal status for dental treatments still unclear. This study aimed to identify porcine DNA in temporary crown, denture base, and hydrocolloid irreversible impression materials using Real-Time PCR. Methods: This research type is descriptive, quota sampling method was used for sampling. Three temporary crown materials, three denture base materials and three irreversible hydrocolloid impression materials were used as the specimens. This study was conducted in two stages. Extraction of DNA sample and Real-Time PCR analysis based on amplification curve and Ct score in yellow and green channel. Results: An analysis of samples based on green channel showed that all materials were not containing porcine DNA, but four out of nine samples were containing vertebrae DNA. Conclusion: All tested specimens were not containing porcine DNA.
